1. Enzymatic Detection
(1) New Trinder’s Reagents (Oxidase system)
New Trinder 的試劑是高水溶性苯胺衍生物 (aniline derivatives),適用於過氧化氫 (hydrogen peroxide)的比色測定。在 4-aminoantipyrine(4-AA) 等偶聯劑 (coupler)存在的情況下,Trinder 試劑會被過氧化物酶 (peroxidase)和兩分子過氧化氫 (hydrogen peroxide)氧化,並呈現紫色或藍色。
(2) Tetrazolium Salts and Electron Mediators (Dehydrogenase system)
四唑鹽 (Tetrazolium Salts)已被開發用於測定脫氫酶活性 (dehydrogenase activity),脫氫酶氧化其底物 (substrate)並還原其輔酶NAD或NADP並產生NADH或NADPH。有色染料是在電子存在下還原 NADH 產生的。為了在水相條件下進行均質測定,水溶性四唑鹽(WST)由於比不溶於水的四唑鹽(如NBT和INT)有更好的水溶性,被廣泛應用於診斷分析和生命科學研究。
